Dipeptidyl peptidase IV deficiency: Its role in peritracheal edema

An experimental study in rodents was designed to assess whether dipeptidyl peptidase IV (DDPIV) deficiency increases peritracheal edema formation during angiotensin-converting enzyme (ACE) inhibition. Magnetic resonance imagery (MRI) was used to determine the volume of ACE-induced edema. Serum DDP activity was decreased in DDPIV deficient (DDPIV-) rats (p<0.001). The volume of peritracheal edema was significantly increased in captopril treated DPPIV- rats (versus DPPIV- saline-treated rats), saline-treated DPPIV+ rats, or captopril-treated rats of the normal substrain (p<0.001). Cotreatment with the neurokinin receptor antagonist spantide attenuated edema in captopril-treated DPPIV- rats (p=0.005 vs. captopril-treated DPPIV- and p=0.57 vs. saline-treated DPPIV- rats). DPPIV deficiency increases peritracheal edema formation when ACE is inhibited through a neurokinin receptor-dependent mechanism (Byrd, ...
